چکیده

The tryptic peptides from human pepsin and gastricsin were purified and their sequences were determined. The sequence of 27 residues at the COOH-terminal end of human pepsin was found to be: -Ile-Leu-Gly-Asp-Val-Phe-Ile-ArgGln-Phe-Tyr-Thr-Val-Phe-Asp-Arg-Ala-Asn-Asn-GlnVal-Gly-Leu-Ala-Pro-Val-Ala. The sequence of 19 residues at the COOH-terminal end of human gastricsin was found to be: -Gln-Phe-Tyr-Thr-Val-Phe-Asp-Arg-AlaAsn-Asn-Lys-Glu-Gly-Leu-Ala-Pro-Val-Ala. These sequences are extensively homologous to each other and to the known COOH-terminal sequence of bovine rennin.

On the specificity of human gastricsin and pepsin.

Digests of oxidized ribonuclease A and glucagon with human gastricsin and pepsin have been compared with IR-120 B column chromatography and two-dimensional high voltage paper electrophoresis to separate the resulting peptides. The amino-terminal residues and the amino acid compositions of purified peptides were determined in order to establish the sequence of peptides and hence the sites of cle...

Specific and irreversible inactivation of pepsin by substrate-like epoxides.

Several substrate-like epoxides were found to act as specific and irreversible inactivators of pepsin. Of these, 1,2-epoxy-3-(p-nitrophenoxy) propane (EPNP) was the most potent. When EPNP reacted with pepsin, all enzyme activity was lost. Two molecules of EPNP were found to be covalently bound to each molecule of completely inactivated enzyme. In this reaction, two carboxyl groups of the enzyme...

Purification and properties of a zymogen from human gastric mucosa.

A previous paper (2) described the isolation and crystallization of gastricsin from human gastric juice and the physical, chemical, and enzymatic properties of gastricsin as compared to human pepsin. It is the purpose of the present report to describe the purification and properties of a zymogen from human gastric mucosa. The activation of this zymogen produces both gastricsin and human pepsin.

Pepsin Inhibitors from Ascaris Zumbricoides

Extracts of the body walls of the adult Ascaris lumbricoides var. suis inhibit pepsin between pH 1 and 6. Four inhibitors of pepsin were isolated and purified as follows. The crude extract of Ascaris was incubated at 37” and pH 2.0 for 75 min. The pepsin-inhibiting activity was obtained in a fraction precipitated at 0.65 saturation with ammonium sulfate at pH 5.35, and sequentially chromatograp...
